nope, using your older sister's given name is unusual in Japan. they just generally use nee-san. Conan uses Ran-nee-chan because he's not related to her, but they're living together, and she's taking care of Conan like a big-sister should. plus, when he was younger Shinichi had a time when he wanted to distance himself from Ran and start calling her Mouri-san instead of Ran, which is more adult, formal, and proper.
